چکیده

M ovement of cargo through the secretory pathway of eukaryotic cells occurs as a discontinuous process involving the activity of 50–80-nm vesicles (28). Vesicles forming on the endoplasmic reticulum (ER) by the COPII coat machinery (35) were first detected as coated elevations on transitional elements directly facing the Golgi apparatus. Subsequently, intermediates harboring ER-derived cargo were found in regions distant from the Golgi stack (16, 33). These structures contained a different coat complex, COPI or coatomer, recognized to be involved in vesicle-mediated recycling (20). It is now apparent that ER export and recycling are tightly coupled. We review recent evidence regarding the organization and role of ER to Golgi intermediates in these events.

Cytoplasmic acidification reduces potassium channel activities in the endoplasmic reticulum of rat hepatocytes

Introduction: Intracellular pH (pHi) regulates essentially all aspects of cellular activities. However, it is unknown how endoplasmic reticulum (ER) potassium channels sense pHi. In this study, we investigate the direct effects of pHi on ER potassium channels. Methods: We used channel incorporation into the bilayer lipid membrane method. L-α-phosphatidylcholine, a membrane lipid, was extrac...

Gating Behavior of Endoplasmic Reticulum Potassium Channels of Rat Hepatocytes in Diabetes

Background: Defects in endoplasmic reticulum homeostasis are common occurrences in different diseases, such as diabetes, in which the function of endoplasmic reticulum is disrupted. It is now well established that ion channels of endoplasmic reticulum membrane have a critical role in endoplasmic reticulum luminal homeostasis. Our previous studies showed the presence of an ATP-sensitive cationic...

Structural organization of the ER–Golgi interface

In the last eighteen months, it has become clear that some classes of proteins are actively recruited into endoplasmic reticulum export carriers, whereas others are exported as bulkflow cargo. Subsequent transport to the Golgi is mediated by tubulovesicular membranes. The anterograde membrane flow is compensated for by a retrograde pathway, which, in addition to the recycling of membrane and pr...

Photoactivation of GFP reveals protein dynamics within the endoplasmic reticulum membrane.

Components of the plant cell secretory pathway, including the endoplasmic reticulum and Golgi apparatus, are in constant motion. The photoactivation of GFP has been used to determine that proteins within the membrane of the ER flow as the ER is remodelled. Measurement of the rate at which activated GFP moves away from the activation spot shows that this motion is much faster than would be expec...

Evidences on the existence of a new potassium channel in the rough endoplasmic reticulum (RER) of rat hepatocytes

Introduction: we have recently reported the presence of two potassium currents with 598 and 368 pS conductance in the rough endoplasmic reticulum (RER) membrane. The 598 pS channel was voltage dependent and ATP sensitive. However, the 368 pS channel was rarely observed and its identity remained obscure. Since cationic channels in intracellular organelles such as mitochondria and RER play imp...

The plant ER-Golgi interface: a highly structured and dynamic membrane complex.

As compared with other eukaryotic cells, plants have developed an endoplasmic reticulum (ER)-Golgi interface with very specific structural characteristics. ER to Golgi and Golgi to ER transport appear not to be dependent on the cytoskeleton, and ER export sites have been found closely associated with Golgi bodies to constitute entire mobile units. However, the molecular machinery involved in me...
